The Eastern Gate was the most important and used gate in the ancient town because it was the one that was leading to Byzantion (and later- Constantinople). 
At the beginning of the new era, the fortress walls of the town were situated closer to the three hills and the Eastern gate itself was situated at another place. Back then, the current location that we see today, was outside of the town walls.
